SIEVEWELL™
Microfludic device for high density single cell capture, culture, processing, and imaging.
Product Basics
SIEVEWELL™ is a slide-sized microfluidic device available in two versions, featuring either 370,000 (20um) or 90,000 (50um) nanowells, each designed to hold a single cell. The specially designed fluidic system helps the user by 1) capturing only one cell in each chamber and 2) maintaining cell position during culture and assays without the need for special equipment. Compatible with cell picking system, making it ideal for single-cell and spheroid isolation.
Key Features
- User-friendly, no specialized equipment needed
- Higher single-cell capture rate compared to a Poisson distribution
- Standard slide glass size, compatible with conventional microscopes
- Ensures stable positioning of suspension cells during live-cell imaging
- SIEVEWELL™ and the cell picking system provide an ideal combination for single-cell and spheroid isolation, with cells securely held during the picking process
Technical Information
Mechanism
Arraying cells is crucial for single-cell analysis, as cell overlap can hinder imaging and isolation. Microcavity arrays are commonly used to capture single cells, with their distribution typically following a Poisson distribution. However, the single cell capture rate tends to be low, particularly when loading large numbers of cells.
SIEVEWELL™ is designed to improve this by achieving a high single-cell capture rate.

Fig 1. Each cell-sized nanowell has 2 pores at the bottom, connected to the side chambers

Fig 2. Aspiration using a pipette from the side port can generate directional liquid flow.

Fig 3. A cell captured in a nanowell blocks the pores to reduce the flow. Free cells will be guided to an empty nanowell.
Applications
- Single cell isolation using micromanipulator
- DNA sequencing
- RNA sequencing
- Cloning
- Spheroid generation from single cells
- On-Chip staining and imaging
SIEVEWELL™ features high-density nanowells, which helps reduce the time needed for imaging.
